      目的 建立了青叶胆中齐墩果酸和熊果酸的反相高效液相色谱-光电二极管阵列检测器(RP-HPLC-PDAD)定量分析方法。方法 采用95%乙醇为溶剂超声提取,色谱柱为Kromasil C18柱(4.6 mm×250 mm,5 μm),柱温30℃。以甲醇:水:磷酸(88:12:0.15,v/v/v)为流动相等度洗脱,流速0.9 mL·min-1,采用光电二极管阵列检测器进行检测,检测波长210 nm。结果 齐墩果酸进样量在0.1048~2.6200 μg时,与峰面积呈良好的线性关系(r=0.9999),平均回收率为96.9%,RSD为1.7%(n=9);熊果酸进样量在0.2304~5.7600 μg时,与峰面积呈良好的线性关系(r=0.9999),平均回收率为97.5%,RSD为1.5%(n=9)。结论 方法准确,操作简便,数据可靠,可用于青叶胆中齐墩果酸和熊果酸的含量测定。

      Objective: A precise and sensitive method for the determination of oleanolic acid and ursolic acid in Swertia mileensis was developed by RP-HPLC-PDAD. Methods: The two compounds were extracted by ultrasonic wave aided method with 95% ethanol solution, and separated on a Kromasil C18 column (5 μm 4.6 mm×250 mm) at 30℃. A isocratic program was carried out at a flow rate of 0.9 mL·min-1using methanol-water-phosphoric acid (88:12:0.15, v/v/v) as the mobile phase. Results: The photodiode array detector was used for qualitative and quantitative analysis and the detection wavelength was set at 210 nm. Oleanolic acid and ursolic acid showed good linear relationship with the peak area (r=0.99999) at the range of 0.1048~2.6200 μg and 0.2304~5.7600 μg, respectively with the respective recovery rates of 96.9% and 97.5%, and RSD of 1.7% (n=9) and 1.5%(n=9). Conclusion: The proposed method is accurate, simple and promises to be applicable for the determination of oleanolic acid and ursolic acid in S. mileensis.
