| 本实验研究建立了金属有机框架材料结合液相色谱,并串联质谱-多肽组学技术分析地龙及其制剂中的多肽成分。首先利用金属有机框架材料NU-1000富集地龙药材及其制剂中的多肽类成分,然后采用液相色谱串联质谱(LC-MS/MS)对富集的多肽成分进行分离分析,并利用De novo软件鉴定其多肽成分结构,再结合地龙数据库对特征肽进行对比,最后采用多肽活性预测软件对其活性进行预测。实验结果表明,利用NU-1000结合LC-MS/MS在地龙中药饮片中鉴定到425条多肽,其中141条肽段来自20个前体蛋白,并有四条多肽FAGDDARP、FAGDDARP、FPSLVGR和FAGDDAARP匹配到了5种蛋白质且均为肌动蛋白,具有抗血栓的药理作用。并在多肽地龙蛋白压片中得到15条肽段具有潜在生物活性的新多肽。本研究建立的地龙及其制剂中多肽成分分析方法操作便捷,可快速分析地龙中多肽成分,有望为地龙多肽类活性成分的挖掘和开发提供技术支持。 |

| A metal-organic framework material combined with liquid chromatography tandem mass spectrometry(LC-MS/MS)-peptideomics technique was established to analyze the peptide components in Pheretima and its preparations. The metal-organic framework material NU-1000 was used to enrich the peptide components in Pheretima and its preparations. The enriched peptide components were separated and analyzed by LC-MS/MS.The structure of the peptides was identified by De novo software, and their characteristic were compared with the Pheretima database. The activity of the peptides was predicted by using peptide activity prediction software. The results showed that 425 peptides were successfully identified in Pheretima by NU-1000 combined with LC-MS/MS. Among them,141 amino acid sequences of peptides derived from 20 precursor proteins, four peptides FAGDDARP, FAGDDARP, FPSLVGR and FAGDDAARP were matched to five proteins and all of them were actin, which had antithrombotic pharmacological effects. A total of 15 new peptides with potential biological activity were obtained from the peptide earthworm protein tablet. The analysis method of the peptide components in Pheretima and their preparations established in this study is easy to operate and can quickly analyze the peptide components, which is expected to provide the support for the mining and developing the active components of Pheretima peptides. |
