Spiders are invertebrates with complex and unique behaviors, making them ideal model organisms for studying animal behavior. The opioid receptor agonist morphine exhibits various neuropsychopharmacological activities, acting on the central nervous system and significantly influencing animal behavior. However, research on the effects of morphine has primarily focused on higher animals, with little attention given to its impact on the behavior of lower animals. The current study aims to investigate how morphine exposure affects locomotor activity and exploratory behavior of the funnel-web spider Pireneitega luctuosa in the open field environment by using the behavioral tracker software. Our results showed that all the spiders, including the untreated, distilled water treated (control) and morphine treated spiders, moved vigorously when they were just introduced into the open field container, then slowed down their speed and became inactive over time. The preference of staying in the peripheral zone of the container over the central zone was also displayed by the spiders. However, the distance of locomotor activity of the spider P. luctuosa was negatively correlated with the morphine exposure concentration within 90 min of the monitoring period. Furthermore, both of the duration and distance of exploration to the central zone dropped down after the spiders being sprayed with the morphine solutions and significant differences being detected between the control and 60 mg/mL-morphine-treated group. Therefore, our study suggests that the morphine exposure may produce a negative effect on the locomotor activity and exploratory behavior in the funnel-web spider P. luctuosa.
