      实验以不同提取方式提取不老莓中黄酮,测定黄酮含量及抗氧化活性得到最佳提取方式。以环磷酰胺(CTX)构建免疫抑制模型,将24只小鼠分为空白组、环磷酰胺组、不老莓提取物低剂量组及不老莓提取物高剂量组。实验开始1 ~ 24 d每日对不老莓提取物低、高剂量组经口灌胃100 mg/mL及200 mg/mL不老莓提取物,空白组、环磷酰胺组灌胃蒸馏水;实验19 ~ 24 d每日对模型组、不老莓提取物低、高剂量组分别腹腔注射80 mg/kg CTX,对空白组注射等体积生理盐水,实验过程中持续观测小鼠体重及采食量,连续注射CTX 5 d后测定小鼠脾脏、肝脏质量及脾脏组织形态,并检测血清中丙二醛(MDA)与超氧化物歧化酶(SOD)含量。实验结果表明:不老莓乙醇提取物黄酮含量最高,不老莓甲醇提取物体外抗氧化活性最佳;与环磷酰胺组相比,不老莓提取物有助于减轻小鼠脾脏、肝脏萎缩,修复小鼠脾脏组织形态学结构损伤,降低血清中MDA含量,提升SOD活性,对CTX所致免疫抑制小鼠具有一定的预保护作用。

      Flavonoids of Rubus vinifera were extracted by different extraction methods, and the contents of flavonoids and its antioxidant activity were determined to optimize the extraction method. The immunosuppressive model was constructed with cyclophosphamide (CTX). 24 mice were divided into blank group, cyclophosphamide group, low dose group and high dose group. At the beginning of the experiment, 100 and 200 mg/mL of the low and high dose groups of R. vinifera extract were orally administered daily from 1 to 24 days, and the blank group and the cyclophosphamide group were given distilled water. From 19 to 24 days, 80 mg/kg CTX was injected intraperitoneally into the model group, low-dose and high-dose extracts groups, respectively, and the same volume of normal saline was injected into the blank group. During the experiment, the body weight and feed intake of the mice were continuously observed. After 5 days of continuous injection of CTX, the spleen, liver weight and spleen tissue morphology were measured, and the contents of malondialdehyde (MDA) and superoxide dismutase (SOD) in serum were detected. The results showed that the ethanol extract had the highest flavonoid contents, and the methanol extract had the best antioxidant activity in vitro. Compared with the cyclophosphamide group, the extract of R. vinifera could reduce the atrophy of spleen and liver in mice, repair the morphological structure damage of spleen tissue in mice, reduce the contents of MDA in serum, increase the activity of SOD, and have a certain protective effect on CTX-induced immunosuppressive mice.
