采用红外辅助提取紫地榆有效成分鞣质,高效液相色谱法同时检测6种鞣质成分含量,以提取时间、料液比、乙醇浓度为自变量进行3因素3水平的中心组合试验。响应面法分析得到优化提取工艺为提取1次,乙醇浓度50 %,提取时间14 min,料液比1:150 g/mL,红外灯275 W,灯距1 cm。经该工艺提取,8批药材平均鞣质总含量为79.06 mg/ g,与预测值80.10 mg/ g相一致(P > 0.05)。优化的红外辅助提取工艺稳定、可靠,为紫地榆总鞣质的有效提取提供理论依据。 |

To optimize the extraction process for total tannin from Geranium strictipes (T-Gs), infrared assisted extraction was employed and coupled with HPLC for simultaneous determination of six compounds of T-Gs. Three-factor, three-level Box-Behnken design was used to optimize some process parameters. The most critical factors affecting the extraction of T-Gs were extraction time, material-to-liquid ratio, ethanol concentration. The optimal infrared extraction conditions for T-Gs were as following: extraction time 14 min, material-to-liquid ratio 1:150 g/mL, ethanol concentration 50 %, infrared lamp height from round bottom flask 1 cm, infrared lamp power 275 W, extraction times 1 time. Under the optimal conditions, the average extraction yield of total tannin from 8 batches of medicinal materials was 79.06 mg/g, which was consistent with the theoretical extraction yield of 80.10 mg/g (P > 0.05). The optimized infrared assisted extraction process for T-Gs is stable and reliable, which provided a theoretical basis for the effective extraction of T-Gs. |
