      采用HPLC法测定橙皮苷、橙皮素单葡萄糖苷和橙皮素在水和不同pH缓冲溶液中的平衡溶解度;结合摇瓶法测定其在正辛醇-水体系和正辛醇-不同pH缓冲溶液体系中的油水分配系数。37 ℃时,橙皮苷、橙皮素单葡萄糖苷和橙皮素在水中的平衡溶解度分别为18.94、632.34、15.37 μg·mL-1;pH值升高,橙皮苷平衡溶解度逐渐降低,对橙皮素单葡萄糖苷和橙皮素无影响,但在pH 7.4时,二者均有最大值,分别为836.57、71.82 μg·mL-1。橙皮苷、橙皮素单葡萄糖苷和橙皮素在正辛醇-水体系中lg P值分别为0.094、0.573、2.876,在不同缓冲溶液中(pH3.4 ~ 7.4)的lg P值范围分别为0.069 ~ 0.281、0.603 ~ 0.664、2.459 ~ 2.905。橙皮素单葡萄糖苷的平衡溶解度远大于橙皮苷和橙皮素,其lg P值在0.6左右,具有一定的亲水亲脂性,预测其在胃肠道中有较好的吸收;橙皮苷的lg P值小于0.3,推测其不易被胃肠吸收;橙皮素的lg P值大于2,其脂溶性极强,易通过主动转运进入生物膜。上述结果可为3个单体成分的成药性,以及中药陈皮中的橙皮苷酶转化为橙皮素单葡萄糖苷的研究,提供实验基础和科学依据。

      The equilibrium solubility of hesperidin, hesperidin-7-O-glucoside and hesperetin in water and different pH buffer solutions was determined by HPLC; the oil-water distribution coefficients in n-octanol-water system and n-octanol-different pH buffer solutions were determined by shaking flask method. At 37 ℃, the equilibrium solubility of hesperidin, hesperidin-7-O-glucoside and hesperetin in water were 18.94, 632.34 and 15.37 μg·mL-1 respectively; with the increase of pH value, the equilibrium solubility of hesperidin gradually decreased, which had no effect on hesperidin-7-O-glucoside and hesperetin, but the maximum values of hesperidin-7-O-glucoside and hesperetin were 836.57 and 71.82 μg·mL-1 respectively, at pH7.4. The lg P values of hesperidin, hesperidin-7-O-glucoside and hesperetin in n-octanol-water system were 0.094, 0.573 and 2.876, respectively. The lg P values of hesperidin, hesperidin-7-O-glucoside and hesperetin in different buffer solutions (pH3.4 to 7.4) were 0.069-0.281, 0.603-0.664 and 2.459-2.905, respectively. The equilibrium solubility of hesperidin-7-O-glucoside was much higher than that of hesperidin and hesperetin, and its lg P value was about 0.6. It has certain hydrophilicity and lipophilicity, which is predicted to have better absorption in the gastrointestinal tract; The lg P value of hesperidin was less than 0.3, so it was speculated that hesperidin was not easy to be absorbed by gastrointestinal tract; hesperetin lg P value was greater than 2, its fat solubility is very strong, easy to enter biofilm by active transport. The above results can provide experimental basis and scientific basis for the study of the medicinal properties of three monomer components and the conversion of hesperidin enzymatic transformation in traditional Chinese medicine Citri Reticulatae Pericarpium to hesperidin-7-O-glucoside.
