      目的 筛选铁苋菜抗炎镇痛作用的有效部位,并初步探究其作用机制。方法 采用热板和醋酸扭体疼痛模型,考察铁苋菜镇痛作用的有效部位,采用二甲苯致小鼠耳肿胀、蛋清致大鼠足跖肿胀以及大鼠棉球肉芽肿炎症模型,考察铁苋菜抗炎作用的有效部位。结果 铁苋菜乙酸乙酯萃取部位明显延长热板所致小鼠痛阈值和明显减少醋酸所致小鼠扭体反应次数,与正常对照组比较和自身给药前比较差异显著(P<0.01);明显抑制二甲苯所引起的小鼠耳肿胀和蛋清所引起的大鼠足跖肿胀以及棉球所引起的肉芽肿,与正常对照组比较差异显著(P<0.01);明显减少醋酸所致疼痛小鼠血清PGE2、cAMP的含量,与正常对照组比较差异显著(P<0.01);明显降低蛋清所致大鼠足跖组织NO、PGE2含量,与正常对照组比较差异显著(P<0.01)。结论 铁苋菜抗炎镇痛作用的有效部位主要为乙酸乙酯部位,其镇痛作用机制可能与降低血清PGE2、cAMP的含量有关,其抗炎作用机制可能与减少炎症组织NO和PGE2含量有关。
英文摘要:
      Objective: To screen the anti-inflammatory and analgesic effective fractions of Acalypha australis, and to study the relative mechanisms. Methods: The analgesic effects of the effective fractions of A. australiswere observed by hot plate and acetic acid writhing model. Meanwhile, the acute inflammatory models, such as xylene-induced ear swelling, egg white-induced paw swelling, and the chronic inflammatory model granuloma induced by cotton pellet implantation, were used in researching the anti-inflammatory effects of the effective fractions of A. australis. Results: After the treatment with low- and high-dose ethyl acetate fraction, the pain threshold with hot plate method was significantly prolonged compared with the control groups and the data before the drugs administration (P < 0.01). The writhing number was reduced after administration compared with the control groups (P < 0.01). Moreover, low- and high-dose ethyl acetate fraction could significantly inhibit ear swelling in mice and paw swelling in rats (P < 0.01). Also, ethyl acetate fraction decreased the granuloma of rats (P < 0.01). The ethyl acetate fraction decreased significantly the serum PGE2,cAMP levels in mice with pain induced by acetic acid,and decreased significantly foot plantar inflammatory tissue NO, PGE2in rat with inflammation caused by egg white (P < 0.01). Conclusion: The ethyl acetate fraction of A. australis is the anti-inflammatory and analgesic effective fractions, and the mechanism of analgesic effect may be related to the reduction of the serum PGE2, cAMP levels, the mechanism of anti-inflammatory effect may be related to the reduction of NO and PGE2 levels in the inflammatory tissue.
