      通过DAVID网站进行上调基因的GO分析,借助STRING工具建立上调基因的蛋白-蛋白相互作用网络图(PPI),使用Cytoscape软件里的MCODE插件筛选出宫颈癌关键基因;借助基因表达谱交互分析数据库(GEPIA)和人蛋白质数据库(HPA)探讨宫颈癌关键基因在宫颈癌组织的表达及预后情况。通过两个数据集获取12个共同上调基因,筛选出TRIP13、ATAD2、POLQ、KIFC1、EXO1、MELK、RMI2、KNTC1、UHRF1这9个关键基因;GO功能富集分析提示宫颈癌关键基因功能的作用主要集中在DNA修复、免疫球蛋白基因的体细胞超突、链置换反应及DNA合成修复等过程中;基因验证结果提示:筛选的9个宫颈癌关键基因在宫颈癌组织的表达均较正常组织高(P<0.05);EXO1、KNTC1、RMI2与患者不良预后相关;免疫组化结果显示KNTC1、RMI2在宫颈鳞癌组织表达较癌旁组织高。本研究通过生物信息学筛选出的与宫颈癌相关的9个关键基因,期待成为宫颈癌诊断和治疗的潜在靶点,并为功能性实验的开展提供理论依据。

      The GO analysis of the up-regulated genes are carried out through the DAVID website, the protein-protein interaction network diagram (PPI) of the up-regulated genes is established with the STRING tool, and the key genes are screened out using the MCODE plug-in in Cytoscape software. Finally, the key genes in cervical cancer tissues are explored by Gene Expression Profile Interactive Ananlysis(GEPIA)and the Human protein database(HPA). Twelve common up-regulated genes are obtained from two data sets, then nine key genes TRIP13, ATAD2, POLQ, KIFC1, EXO1, MELK, RMI2, KNTC1, UHRF1 are screened out. GO function enrichment analysis suggests the main functions are mainly concentrated in the DNA repair process, the somatic hypermutation process of immunoglobulin genes, strand replacement reaction, DNA synthesis repair, and so on. The verification results show that the expression of nine key genes in cervical cancer tissues is higher than that in normal tissues (P < 0.05), and EXO1, KNTC1, and RMI2 are related to the poor prognosis of patients. The immunohistochemical results of KNTC1 and RMI2 proteins recorded in the database suggest that the expression level of cervical squamous cell carcinoma tissues is higher than that of adjacent tissues. The nine key genes related to cervical cancer screened out in this study through bioinformatics may be the potential targets for treatment and provide a theoretical basis for the development of functional experiments.
