      通过MTT法、细胞粘附试验、Transwell细胞迁移和侵袭试验检测不同浓度的大蒜素对肺腺癌细胞的活性、粘附、迁移与侵袭能力的变化;(RT-qPCR)逆转录-定量聚合酶链反应检测不同浓度的大蒜素对TIMP/MMP平衡的影响。本研究中发现大蒜素可呈剂量依赖性抑制肺腺癌细胞的活性、粘附、迁移和侵袭能力。大蒜素主要降低基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的mRNA水平,同时对于组织金属蛋白酶抑制剂-1(TIMP-1)和组织金属蛋白酶抑制剂-2(TIMP-2)的mRNA水平则相反地呈剂量依赖性增加。因此我们得出结论,大蒜素可能通过抑制TIMP/MMP平衡,从而抑制肺腺癌细胞的侵袭。大蒜素可作为肺腺癌治疗的潜在抗侵袭剂。
英文摘要:
      MTT method, cell adhesion test, Transwell cell migration and invasion test were used to detect the changes in the activation, adhesion, migration and invasion of lung adenocarcinoma cells at different concentrations of allicin; (RT-qPCR) reverse transcription-quantitative polymerization enzyme chain reaction was used to detect the influence of different concentrations of allicin on the balance of TIMP/MMP. It was found that allicin could inhibit the activation, adhesion, migration and invasion of lung adenocarcinoma cells in a dose-dependent manner. Allicin mainly reduced the mRNA levels of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9), and at the same time for tissue metalloproteinase inhibitor-1 (TIMP-1) and tissue metalloproteinase inhibitor- 2 (TIMP-2) mRNA levels increased in a dose-dependent manner on the contrary. It could be concluded that allicin may inhibit the invasion of lung adenocarcinoma cells by inhibiting the balance of TIMP/MMP. Allicin could be used as a potential anti-invasive agent in the treatment of lung adenocarcinoma.
