目的 基于BATMAN-TCM在线分析工具研究钩藤主要活性成分及药理机制。方法 从BATMAN-TCM数据库中提取钩藤的化学成分,以Score cutoff ≥ 30和P ≤ 0.05筛选活性成分及作用靶点,分析靶点富集的信号通路与疾病,构建活性成分-靶点-通路-疾病网络,预测钩藤的多维药理机制。结果 获得钩藤碱、毛钩藤碱、卡丹宾碱等23个主要活性成分;以及DRD2(多巴胺受体D2基因)、HTR1A (5-羟色胺受体1A)、HTR1B (5-羟色胺受体1B)、SLC18A2等21个核心靶点;活性成分可关联细胞间隙连接、神经活性配体-受体相互作用、钙信号通路等通路11条;可治疗疼痛、心血管系统和神经系统等相关疾病21种。可进一步验证钩藤"多成分-多靶点-多通路"的作用机制。结论 本研究从网络药理学角度探讨钩藤主要活性成分、相关通路及多种药理作用机制,为进一步探讨其作用机制奠定了基础。 |

Objective: To study the main active ingredients and pharmacological mechanism of hookvine from the perspective of network pharmacology.Methods: Chemical constituents of hookvine were extracted from BATMAN-TCM database.Score cutoff ≥ 30 and P ≤ 0.05 were used to screen active constituents and action targets.Signal pathways and diseases of target enrichment were analyzed.Active components-targetpathway-disease network was constructed to predict the multidimensional pharmacological mechanism of hookvine.Results: 23 major active components such as rhynchophylline,irradiata,and kadanbin base were obtained.These components can act on 21 core targets such as DRD2(dopamine receptor D2),HTR1A (serotonin receptor 1A),HTR1B (serotonin receptor 1B),SLC18A2,etc;active ingredients can associat with 11 pathways such as cell gap junction,neuroactive ligand-receptor interaction,and calium signaling pathway;it can treat 21 related diseases such as pain,cardiovascular system and nervous system.The mechanism of "multi-component-multi-target-multi-pathway" of hook-vine can be further verified.Conclusion: In this study,the main active components,related pathways and various pharmacological mechanisms of hookvine were discussed from the perspective of network pharmacology,which laid a foundation for further exploration of its mechanism of action. |
