      目的 分离、纯化及鉴定消癥益肝片中抗肝癌的活性物质。方法 采用Sephadex LH20和反相制备高效液相色谱柱分离纯化;采用MS、1H-NMR等波谱分析进行结构鉴定;利用MTT法检测化合物的抗肝癌活性。结果 经分离纯化,获得3个活性小分子化合物,分别为苯甲酸(I)、对羟基苯甲酸乙酯(Ⅱ)、对羟基苯甲酸丙酯(Ⅲ)。MTT实验表明,三个化合物均有体外抑制HepG2细胞增殖的活性,其中化合物Ⅱ和Ⅲ作用较强,并呈时间-剂量依赖关系。结论 鉴定出3个小分子化合物,均为首次从消癥益肝片中分离得到,具有体外抑制肝癌细胞生长的活性。
英文摘要:
      Objective:To isolate, purify and identify the anti-hepatoma constituents from Xiaozhengyigan tablets (XZYG). Method:Anti-hepatoma chemical constituents were isolated and purified by Sephadex LH-20 and preparative RP-HPLC. The chemical structures were identified by MS and 1H-NMR. The effect on the proliferation of human liver cancer cell line (HepG2) was evaluated by MTT assay. Results:Three chemical constituents were obtained and their structures were determined as benzoic acid (I), Ethyl p-hydroxybenzoate (Ⅱ), Propyl-p-hydroxybenzoate (Ⅲ), respectively, based on MS and 1H-NMR data and comparison with the literatures. All of the three compounds were isolated from XZYG for the first time and had growth inhibitory activity on HepG2 cells. The inhibitory effect of Ⅱ and Ⅲ on HepG2 cells showed a concentration-dependent manner. Conclusion:Three small molecules were isolated from XZYG which may inhibit tumor cell growth in vitro.
