      目的 采用大鼠大强度跑台运动模型,以人参作为阳性对照,探讨不同剂量螺旋藻补充对运动诱导的大鼠海马形态结构损伤及脑源性神经营养因子(BDNF)海马区表达的影响。方法 将50只SD大鼠随机分为正常对照组(NC组)、运动模型组(EM组)、运动+螺旋藻灌胃组(ESⅠ组)、运动+螺旋藻高剂量组(ESⅡ组)和阳性对照组(PC组)。每组10只。观察螺旋藻低、高剂量补充对运动大鼠体重等一般情况、血清LDH活性和BUN含量、海马CA1区微细形态结构和BDNF表达的影响。结果 (1)实验第三周末,ESⅠ组和ESⅡ组疲劳相关症状得到减轻,大鼠食欲和体重增加。(2)与EM组比较,ESⅡ组大鼠血清LDH活性增加和BUN含量明显降低(均P<0.01)且较PC组已无明显差异(P>0.05)。(3)EM组海马CA1区神经元数目、胞浆尼氏小体数目明显减少;而ESⅠ组和ESⅡ组海马神经元损伤均较EM组明显减轻,神经元数目、尼氏小体数量逐渐增多。(4)与EM组比较,ESⅠ组和ESⅡ组大鼠海马BDNF蛋白表达均增加,但ESⅡ组增加更为明显(P<0.01);与PC组比较,ESⅡ组和ESⅠ组BDNF蛋白表达均降低,但ESⅠ组降低更为明显(P<0.01)。结论 螺旋藻低、高剂量补充可改善运动所致的海马形态结构损伤,其中高剂量组的改善效果要优于低剂量组,其原因可能与高剂量螺旋藻补充能较好的上调海马区BDNF蛋白表达,进而对运动大鼠表现出明显的神经保护作用有关。
英文摘要:
      Objective:To investigate the effects of different dose spirulina on morphological structure injury of hippocampal and BDNF expression in the exercise rats through the high-intensity treadmill exercise model using Ginseng as a positive control. Methods:The SD rats were randomly divided into normal control group(NC), exercise model group(EM), exercise plus low dose group(ESⅠ), exercise plus high dose group(ESⅡ)and positive control group(PC), 10 rats each group. Body weight, serum LDH activity, serum BUN content and BDNF expression were tested, and micromorphology changes of hippocampal CA1 were observed by light microscope after the experiment end. Results:Group ESⅠand Group ESⅡshowed significantly lessened symptoms related to fatigue, and increased appetite and body weight compared to Group EM after the 3th week. (2) Serum LDH activity of Group ESⅡwas higher and BUN content was lower than those of Group EM(P<0.01), and there was no significant difference from Group PC. (3) The number of neurons and Nissl bodies were significantly decreased in the hippocampal CA1 of Group EM, while hippocampal neurons injury in Group ESⅠand Group ESⅡwere significantly reduced compared with Group EM. BDNF expressions in Group ESⅠand Group ESⅡwere increased compared with Group EM, and Group ESⅡwas more increased. The protein expressions of Group ESⅠand Group ESⅡwere lower than those of Group PC, while the expressions of Group ESⅡ was higher than those of Group ESⅠ(P<0.01). Conclusion:Low and high dose spirulina supplement could improve morphological structure injury of hippocampal, and the improvement of high dose group was better than that of low dose group. That may be related to upregulating BDNF expression with neuroprotective effects in the hippocampus of exercise rats.
