      采用响应面法对酶解提取鳊鱼鱼鳞胶原蛋白的工艺进行优化。在单因素实验的基础上,根据Box-Behnken实验设计的原理,以酶解温度、pH值、酶解时间为自变量,以胶原蛋白含量为响应值进行响应面优化分析。结果表明:设定的3个影响鱼鳞胶原蛋白提取效果的因素,其影响大小依次为pH值 > 酶解温度 > 酶解时间;优化分析获得鱼鳞胶原蛋白的最佳工艺条件为:酶解温度31.32℃、pH值4.24、酶解时间1.95 h、加酶量1.5%,胶原蛋白的含量为883.64 mg/g。为了方便实际操作,将鱼鳞胶原蛋白的最优工艺条件修正为:酶解温度31℃,pH值4.2,提取时间2 h、加酶量1.5%,实际得到胶原蛋白含量为881.78 mg/g,实验结果与理论值基本吻合,说明模型的可靠性较高。

      Enzymolysis in extraction process of bream fish scale collagen was optimized by response surface method. On the basis of single factor experiment, taking enzymolysis temperature, pH value and enzymolysis time as independent variables, the content of collagen as the response value, the response surface method was applied to optimize according to the principle of Box-Behnken experimental design. The results show that the effects of three factors influencing scale collagen extraction are:pH value > enzymolysis temperature > enzymolysis time. The optimal process conditions achieved by optimization analysis for bream fish scale collagen are:temperature 31.32℃, pH value 4.24, enzymolysis time 1.95 h, and enzyme addition 1.5%. The collagen content is 883.64 mg/g. For the sake of convenience in practical operation, the optimal process conditions of fish scale collagen are modified as enzymolysis temperature 31℃, pH value 4.2, extraction time 2 h, and enzyme addition 1.5%. The actual collagen content is 881.78mg/g. The results agree with the theoretical value, which shows the high reliability of the model.
