      观察油橄榄叶提取物(OLE)对非酒精性脂肪肝(NAFLD)的防治作用。用高脂饮食9周建立小鼠NAFLD模型,造模第2周起用OLE(250,500,1000 mg/kg)进行灌胃治疗8周,利用生物显微技术观察肝脏组织结构的变化,全自动生化分析仪检测血清甘油三酯(TG)、胆固醇(CHOL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,用放射免疫法检测肝脏肿瘤坏死因子α(TNF-α)、白细胞介素1(IL-1β)、白细胞介素10(IL-10)含量,比色法检测肝脏超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽还原酶(GR)活性及丙二醛(MDA)含量。研究结果显示,与模型组比较,OLE治疗后,血清TG、CHOL、ALT、AST水平降低;肝脏TNF-α、IL-1β、MDA的含量显著降低,IL-10、SOD、CAT、GR水平均显著升高,肝组织损伤明显减轻。OLE可改善肝脏脂质代谢,减少自由基损伤,缓解肝脏脂肪变性和炎症因子反应,抑制NAFLD的进展。

      The aim of study was to explore the protection role of olive leaf extract (OLE) on nonalcoholic fatty liver disease (NAFLD). The mouse NAFLD model was induced with high fat diets for nine weeks. Since the second week after modeling, the mice were given OLE at doses of 250, 500 or 1 000 mg/kg daily by intragastric administration for eight weeks. The changes of liver histological structure were observed by bio-microscopy. The levels of triglyceride (TG), cholesterol (CHOL), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum were tested by automatic biochemistry analyzer. The contents of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-10 (IL-10) in liver were detected by radioimmunoassay. Spectrophotometry was used to detect the activities of superoxide dismutase (SOD) and catalase (CAT), glutathione reductase (GR) and malondialdehyde (MDA) content in liver. The results showed that compared with the model group, after the treatments of OLE, the levels of TG, CHOL, ALT, AST, TNF-α, IL-1β and MDA were significantly decreased, the levels of IL-10, SOD, CAT and GR were significantly increased. Further experiments found that OLE decreased the liver injury. The results indicated that OLE can improve the lipid metabolism, reduce free radical lesion, relieve fatty degeneration and inflammatory response, and prevent NAFLD occurrence and development.
