      目的 观察升清降浊方对CRF大鼠的作用机制。方法 SD大鼠160只,随机分为4组:即空白组,肾衰模型组,阳性药物对照组,升清降浊方组,每组40只。上述大鼠制作CRF模型,按相应组方给药28 d,采血后处死,取肾组织进行HE染色和电镜观察病理变化;采用全自动生化检测仪,对收集的血清样本进行肌酐(Scr)、尿素氮(Bun)和尿酸(UA)等生化指标检测,并进行用药前后数据对比;采用实时荧光定量PCR方法比较各实验组不同用药前后血液及给药后肾组织中的白细胞介素1(IL-1)、白细胞介素6(IL-6)和肿瘤坏死因子(TNF-a)基因的mRNA表达水平差异,并取肾组织采用免疫组化和Western-blotting法测定大鼠肾IL-1、IL-6和TNF-a蛋白表达。结果 升清降浊方组与空白组的IL-1、IL-6和TNF在两组间统计学检验情况比较,IL-1,IL-6实验组显著高于对照组,TNF实验组显著低于对照组,比值明显下降(P<0.05);升清降浊方组与肾衰模型组、阳性药物对照组的肾功能指标情况存在显著性差异有统计学意义(P<0.01);肾组织HE染色及IL-1、IL-6、TNF-a、免疫组化染色升清降浊方组与空白组、肾衰模型组、阳性药物对照组均间存在显著性差异。结论 升清降浊方通过调控炎症因子IL-1、IL-6和TNF-α蛋白表达,从而阻断病情进展,改善肾功能。
英文摘要:
      Objective: To observe the mechanism of ascending qing and descending zhuo prescription in CRF rats.Methods: 160 SD rats are randomized into 4 groups---blank group,renal failure model group,positive drug group and ascending qing and descending zhuo prescription group;each group contains 40 SD rats.These rats are used to make CRF model,and killed for blood collection after administration for 28 days according to the corresponding formula;HE staining is used in renal tissue to observe its pathological changes by electron microscope.Based on serum samples,biochemical indexes such as serum creatinine (Scr),blood urea nitrogen (Bun) and uric acid (UA) are tested by automatic biochemistry analyzer and the data before and after medication are compared.Real-time quantitative PCR method is used to compare mRAN differences in the genes of interleukin 1(IL-1),interleukin 6(IL-6) and tumor necrosis factor (TNF-a) in blood before and after medication and renal tissue after treatment of each group;and immunohistochemistry and western-blotting method are used to test the expression of IL-1,IL-6 and TNF-a proteins in renal tissue.Results: After analyzing the statistical tests of IL-1,IL-6 and TNF between ascending qing and descending zhuo prescription group and blank group,IL-1 and IL-6 in experimental group are significantly higher than those in the control group,but TNF is significantly lower and the ratio decreases obviously (P<0.05);there is significant difference in kidney function in ascending qing and descending zhuo prescription group,renal failure model group and positive drug group(P<0.01)and in renal tissue HE staining,IL-1,IL-6,TNF-a and immunohistochemical staining in all the groups.Couclusion: Ascending qing and descending zhuo prescription blocks the progression of the disease,and improves the function of kidney through regulating protein expression of inflammatory factors IL-1,IL-6 and TNF-α.
