      探讨17β-雌二醇(E2)和17α-甲基睾酮(MT)暴露对食蚊鱼(Gambusia affinisCYP19a基因表达的影响。采用静水暴露方法,分别设置2、10、50、100 ng/L E2和MT各4个实验组,并设置对照组和平行组,定量测定第7,14,21,28 d性腺组织CYP19a mRNA表达水平的变化。E2暴露实验结果显示,初始相对较低浓度的E2(2、10 ng/L)对食蚊鱼CYP19a基因的表达起促进作用,而较高浓度的E2(50、100 ng/L)对CYP19a起抑制作用;随着暴露时间的延续,食蚊鱼CYP19a基因的表达受低浓度E2的影响不明显,而高浓度的E2则极明显上调食蚊鱼CYP19a基因的表达。MT暴露的实验结果显示,初始所有MT实验浓度组(2、10、50、100 ng/L)均对食蚊鱼CYP19a基因起促进作用;随着暴露时间的延长,较低浓度MT对食蚊鱼CYP19a基因产生的影响不明显,较高浓度的MT则显著性地抑制食蚊鱼CYP19a的表达。结果表明,两种典型环境激素对食蚊鱼CYP19a基因表达的影响十分显著,鱼类CYP19a基因mRNA表达适合作为环境激素污染的生物标志物。

      The effects of 17β-estradiol (E2) and 17α-methyl testosterone (MT) exposure on CYP19a gene expression in Gambusia affinis were investigated in this paper. G.affinis were randomly divided into five groups, with one control group and four experimental groups (2, 10, 50 and 100 ng/L E2 and MT, respectively). Parallel experimental groups were also established. By using the hydrostatic exposure method, the CYP19a mRNA expression levels in fish gonad tissues were determined after 7, 14, 21, and 28 d exposure, respectively. E2 exposed experimental results showed that, the initial, relatively low concentration of E2 (2 and 10 ng/L) promoted the G.affinis CYP19a gene expression level, while the high concentration of E2 (50 and 100 ng/L) inhibited the CYP19a gene expression. With the extension of exposure time, the expression of CYP19a gene was not significantly affected by low concentrations of E2, while high concentrations of E2 were significantly up-regulated in the expression of CYP19a gene. MT exposed experimental results showed that, the initial, all the MT experimental concentrations (2, 10, 50 and 100 ng/L) had promoted the G.affinis CYP19a gene expression level, with the extension of exposure time, effects of low concentration of MT on the production of CYP19a gene expression were not obvious, while the high concentrations of MT significantly inhibited the CYP19a gene expression. The results indicated that, the effects of two typical environmental hormones on G.affinis CYP19a expression were very significant, fish CYP19a gene mRNA expression was suitable as a biomarker for environmental hormone pollution.
